The $5,000 mark is well-known in the Signature circles of Visa. But how about American Express? If you transfer credit limits among your Amex cards and one card is lowered below $5,000, is it significant in any way to anyone?

It also seems to be turning ino myth as not all Signatures have a $5K requirement. AmEx has no tiers defined by a $5K limit.
No, Amex couldn't care less.

This does not apply to American or Discover. It's just a Visa and MC benefit.
Visa and MC draw that slighttly-fuzzy line at $5k because their interchange fees are lower than Amex's. To provide and pay for the Signature/WEMC level of card benefits, they want to know that you potentially can (and hopefully will) put a high level of spend through the card.
Since Amex has a higher fee, they really don't need to draw that line.
Also, since Amex is both the network and the lender, they can take a more holistic view of these matters.
Amex is much more sensitive to how your total limit relates to your total income than where any particular single card's limit is set.
