Usually, you need to call the number that is on the sticker attached to the front of your new card.
You will usually need to call from the same telephone number that they have have on file from your application.
This is done so that their "caller ID" can verify that the card is actually in your possession.
The automated activation prompts (or sometimes an actual human representative) will ask you to verify the card number and other information - in doing this, they are able to verify that the card was not lost of stolen from the mail, and that you are in fact, the person who applied for the card.
Until the card has been activated, it doesn't technically "exist", and can not be used, and any attempts to swipe it will simply not be recognized by the issuer - and it might even result in the card being immediately cancelled as a stolen card, requiring you to call customer service to explain the matter.
Now they give you the option to activate online. I just activated one yesterday but it was a renewal, not a new account. Either way, you'll have to verify some info.
Congrats on your new card. Capital One activation process is painless even for over the phone activation. Glad to hear that they offer online activation now.
