AMEX CLI

If you apply for CLI and are denied, how long do you have to wait, if at all, before requesting an increase again?

Depends on if you're talking about the 1st one ever on the account.  If it's the 1st one and you're not 61 days plus then when it's day 62 hit the button again.  If it's any other situation then it's 90 days on a denial and 180 days for approved ones.

Was your denial due to asking too soon or due to credit?  Do you have other Amex cards?  Is this a new card or have your received CLI's on this card previously?

While your specific question has been answered, in the future, I'd suggest you take a look at the linked thread.  While very long, the inital post contains a wealth of info about Amex CLIs and it will always be beneficial to occasionally take a look at some of the recent posts to get an idea of what's going on with CLI requests
