Does Chase Update automated line on weekends?

As soon as I'm approved via the automated line, after verification process, can I call at that time and ask for the card to be overnighted? Business trip next Sunday. Would like to start my spend for that bonus.

You can ask but if you're talking about the Marriott card it won't be overnighted  immediately after approval.  Cards like the Ritz, Marriott and Hyatt take extra time because they have to link the cards with the hotel rewards account.  Usually you will receive those cards in the 7-10 day period after approval.

Yeah I've read that, so I didn't even try after that approval, although my status on Marriott Rewards site, was upgraded to silver already. So seems like the card must be linked to my rewards account already. My second app, CSP went to 2 weeks message last week, after DOB verification was received I suspect, from the 7-10 day message. I was wondering on that one.

Usually the 2 week message means an approval but Chase can be difficult to gauge for sure.  They will overnight the CSP so when you're approved, call them up.
