OP, you paid this off WAY too fast.
There is a slow reveal going on here, likely some more relevant facts that would be helpful to really understand the full situation.
You should have no problem once they get the bank account and a hooman reviews your account. The initial reaction was by a computer algorithm, likely watching for money laundering.
I would guess PayPal is not the primary issue here, or elsewhere on AMEX charges, but rather other aspects of the use of PayPal:
1) Purchasing from family or linkable friends
2) Buying from yourself
3) Paying way more than the posted balance on your account when they don't even have the visibility whether 1 or 2 above applies yet.

+1 on the slow reveal. There seems to be more to this story than we are hearing.
Hopefully $5000 in "business supplies" wasn't beakers and chemistry equipment to make some blue crystal, lol

That's why I was asking about that. I've seen a handful of posts about an overpayment (since the purchase hadn't even posted as a fully completed charge yet, your payment was likely causing your "current balance" to be thousands in the negative) triggering some fraud alerts with Amex. Or it could have been something that triggered a money laundering alert with Amex as Pooka said. Amex seems to highly prefer paying after your statement has cut and paying in full prior to the due date.
