There's a tab on my AMEX site that says Recent activity and it pretty much adds up the total amount I've spent since the last closing date. In this case $581.

Yes they will look at your recent activity for the full month.
They will also look at your average balance.
They're not going to "match" anything, but some people have reported that by having a lot of activity in an Amex credit account, it helped them get a CLI, but I wouldn't count on it if I were you.
I've been with Amex for over a year and a half and have never gotten a CLI on either of my credit card accounts.
