Most of them just do the legwork for you as far as travel arrangements and restaurant reservations and so on, but don't really have any pull.
I'm pretty sure the concierge service that comes with Amex Platinum does have some pull, however, and can get a table at a restaurant that's booked for the night. Just as an example.
Edit to add: Look at all the high-roller-type discussions we have going on tonight! First lounges, now concierge services!

I haven't used the services before, but I was with my former employer on a couple of occasions when he used the service with his Amex Platinum. It was pretty impressive. He had a client visiting from out of town and was able to get us into Morton's Steakhouse in Chicago at the very last minute, after the restaurant had told him that getting a table that night would be "highly unlikely."

I do anticipate that I'll be using the service on my Citi Platinum Amex pretty frequently, though. Their first test will be to see what kind of seats they can score me when my Cubs come to town to play the Mets in September. 
For the record, I don't have an Amex Platinum. Just got the Citi Platinum Amex.
