If you've been PIF and your scores from your approval and current reported one from AMEX website are basically the same you should be good to go in requesting. It won't hurt to pay it off first before requesting but don't think it's necessary.
If I were in your shoes I'd make the request. Its only a soft pull and the worst that could happen is that Amex will say no.

Well you sufficiently scared me with your comments about the cash advance, so I paid the full balance today & am waiting for it to post before I submit the CLI. I will update as soon as I know.
