So my last "automatic" 2.5X CLI from Amex was April 10. When do you think I should ask for another CLI? online or call?

My understanding is that the auto CLI's don't reset the time. So I believe you could apply 6 months from your last approval, although I have been seeing reports of people applying 3 months from last approval and being approved.
But wait to hear from an Amex CLI expert.

I'm by no means an expert, however I have never had a bad result when I wait 6 months with Amex... @SouthJamaica is correct about auto increases not 'always' resetting the clock. I've never been done wrong by them waiting for 6 months. It's my personal rule with them.
what do you guys mean by "Not resetting" the clock? Does it mean that if I applied let's say in January, regardless of the outcome, if I get an automatic CLI in April, then by July, it would be 6 months since my request of a CLI?
If you ask for a CLI and get one. Its 6 months until you can ask again. If you get an auto CLI. You can ask anytime. I got a auto CLI a couple months ago. Next day I asked for a CLI and got it.

The rule used to be (and I don't know that it still is) that you had to wait 6 months after a CLI approval, and 3 months from a CLI denial, but that an auto CLI didn't count in the computation; i.e. the rule only applied to CLI requests. So an auto CLI, since it hadn't been requested, didn't "reset" the 6 month/3 month "clock".

So I went ahead now and requested an increase. When I opened the account last year, I was given $1,000. The auto CLI gave me a CL of $2,500. So I asked for $7,500. BAM! Approved. Do you think had I asked for more, I would have been denied?

Congratulations. You're on a roll.
Yes I'm pretty sure that had you asked for more you would have been denied. The CLI you received -- going from 2500 to 7500 -- is Amex's classic "3X CLI", although I don't know why they call it that since the CLI part is really 2X the previous limit.
Now you wait. The big question is how long. Historically, you would have to wait 6 months until your next CLI application. But I think I may have seen reports of people getting CLI's sooner. You'll have to wait for more serious Amex CLI-watchers to chime in on this. I haven't asked Amex for a CLI in ages, because my limits are way higher than my spending warrants.
